• 非天然アミノ酸の導入のための遺伝コードの拡張

• 生体内のタンパク質合成では、遺伝子上の三つ並びの核酸塩基(コドン)が、遺伝暗号表で決められた１つのアミノ酸へ翻訳されている。私達はコドンを四つ並びに拡張した「４塩基コドン」を用いて、天然には存在しないアミノ酸「非天然アミノ酸」をタンパク質の特定部位へ導入することのできる新たなタンパク質合成システムの開発に成功している。このシステムを基礎として、以下に示す新しいタンパク質改変技術の開発とその応用展開を試みている。 ・蛍光標識アミノ酸の導入によるタンパク質の構造機能解析技術の開発 ・非天然アミノ酸導入技術の高効率化 ・修飾アミノ酸のタンパク質への導入技術の開発 ・部位特異的変異導入のための新技術の開発研究課題
